I like Rob Wells and TPB but they don't even come close to entering the "funniest ever" discussion..
Maybe not, but they are funnier than Vince Vaughn

Considering this question is as subjective as asking who is the most beautiful woman of all time....

This is far too hard of a question.
I loved Hedberg when he was alive- got to see him- admired his writing... but was he really a funny person?... Oddly enough, I'd say no. He was brilliant, but when I think funny people... he's more creative than "hilarious". Much like Demetri Martin is.
I think Bo Burnham is the best current stand-up comedian. He combines a bit of those elements, but has a bit more "funny guy" personality with it.
Richard Pryor, Steve Martin, Eddie Murphy (in his prime- untouchable), Dave Chappelle... those guys I see as more funny people.
Hell, I'll probably catch hate for it- but Kevin Hart fits that mold too. Dude stays busy, and he's always funny.
Then you've got guys that just have amazing comedic timing on screen- Will Arnette, Jake Johnson, David Cross, Red Fox, Gene Wilder, etc.
